چکیده

The considerations which prompted an investigation of the r61e of folic acid in formate metabolism were discussed in an earlier paper (1) in which it was reported that rats deficient in this vitamin incorporated less of a standard dose of CY4-formate into certain amino acids and heme than did rats fed folic acid. There is abundant microbiological evidence to indicate an interrelationship between folic acid and the purines (2-5). The accumulation of 4-aminoimidazole-5-carboxamide in cultures of Escherichiu coli inhibited with sulfanilamide (6) suggests very strongly that folic acid controls the incorporation of formate at least into carbon atom 2 of the purines. In addition, Skipper et ab. (7) have demonstrated that the incorporation of labeled formate into the total nucleoprotein of mice is decreased by the administration of folic acid antagonists. This study was undertaken to demonstrate whether folic acid affects the incorporation and relative location of labeled formate in nucleic acid purines in the rat.

Effects of a folic acid antagonist on nucleic acid metabolism.

Microbiological evidence (1) first implicated folic acid in nucleic acid synthesis, and several lines of investigation suggest that citrovorum factor, or another derivative of folic acid, may function in some manner as a coenzyme in the metabolism of a l-carbon moiety. The function of the formy1 group in rhizopterin (2) and citrovorum factor (3) has not been clarified. The incorporation of form...

GAR transformylase, which is responsible for the incorporation of formate carbon into what becomes position 8 of the purine ring, has been completely separated from 5-amino-4-imidazolecarboxamide ribotide transformylase (3). The latter enzyme is responsible for the incorporation of formate carbon into position 2 of the purine ring. The requirement for folic acid compounds has been demonstrated ...

The relationship of folic acid to formate metabolism in the rat.

There are many indications that vitamins containing p-aminobenzoic acid (PABA) are concerned with the synthesis of a variety of metabolites (l-16). The mode of action of the PABA-containing vitamins is suggested by the work of Shive et al. (ll), who found that the amine which accumulates in sulfanilamide-treated Escherichiu coli (12) is 5-amino-4-imidazolecarboxamide. The addition of only a sin...

Biosynthesis of the purines. III. Reactions of formate and inosinic acid and an effect of the citrovorum factor.

Previous experiments have shown that when hypoxanthine is synthesized de novo from formate, glycine, and CO2 in extracts or homogenates of pigeon liver these substrates are utilized for purine synthesis in the molecular ratio of 2: 1: 1 (1). Formate is the precursor of carbon atoms 2 and 8 of the purine base; glycine, of atoms 4, 5, and 7; and CO2 of atom 6 (2, 3). The present problem originate...
